研究概要	高精度CGH(comparative genomic hybridization)アレイを開発し、遺伝疾患や癌細胞に起きた微細ゲノム構造異常の探索を染色体ワイドに実施して、検出した疾患特異的ゲノム構造異常を糸口に、疾患の発症に関連する遺伝子を同定することを目的に研究を進め、以下の成果が得られた。 1.総数900種を超える種々の癌においてゲノムコピー数異常を解析しデータベースを構築した。(http://www.cghtmd.jp/) 2.新規の遺伝子増幅領域を探索し、食道扁平上皮癌のcIAP1,ZASC1、神経芽細胞腫のPPM1Dをはじめとする種々の標的癌関連遺伝子を明らかにした。 3.総数5,535個のBACクローンを搭載した高密度、高精度ゲノムアレイを構築した。これを用いて、微細ヘミ欠失を検出することが可能になっており、従来、本態不明とされてきた遺伝的背景の強い疾患群において、疾患遺伝子同定の糸口となるゲノム構造異常を特定することに期待がかかる。 4.ゲノムアレイの応用により、メチル化DNA領域の網羅的検出を可能とするシステムを構築した。これにより、神経芽細胞腫において分化のスイッチのON/OFFに関与する可能性のある遺伝子が同定されてきている。 5.ゲノムアレイで、従来では明らかにされることの無かった新規のホモ欠失領域の複数を、種々の癌に見出だし、同領域より癌抑制遺伝子候補を特定しつつある。
