研究概要

TLR経路は生体防御に必須であるが、当該研究にて、肝臓の恒常性維持ならびに急性関節炎においても重要な役割を果たすことが判明した。
(1)TLRを介したシグナルは、種々の制御分子により抑制される。サイトカイン制御因子であるSOCS-1もTLRシグナルに対して負の制御を行う。ところで、当該研究で、TLRを介したシグナルが、肝細胞のホメオスターシスに必須であることを明らかにした。事実、TLRシグナルに必須のシグナルアダプター分子であるMyD88を欠損したマウスは、部分肝切除後の肝再生が不良で、肝再生に必須の再生誘導因子の発現も欠如していた。SOCS-1組み込んだベクターを投与して、このTLRシグナル依存性の肝再生プロセスに過剰なSOCS-1がどのような役割を果たすかを検討したところ、過剰なSOCS-1を発現したマウスは、肝再生不良であることが判明した。以上の結果から、TLRシグナルは臓器ホメオスターシスに必須で、SOCS-1はそのシグナルを制御することが判明した(Hepatology,41:443,2005)。
(2)マウスはLPS耐性である。しかし、事前に大腸菌成分を投与すると、LPSの関節腔内注射によって重篤な急性関節炎が惹起される。この関節炎は、細胞内シグナルアダプター分子であるMyD88依存性である。通常、炎症性関節炎はTNFの中和により減弱することがよく知られているが、当該関節炎はTNFではなくIL-6をブロックすることで、完全な予防が可能であった。
以上の知見は、TLRを介する自然免疫応答が、生体防御のみ成らず、多様な生物現象に深く関わることを如実に示すものである。
