アディポネクチンを用いた炎症性腸疾患の治療法の開発

研究概要	1)炎症性性腸疾患におけるアディポネクチンの発現動態の検討(船橋) クローン病患者におけるアディポネクチンの発現および産生動態を検討した。クローン病患者の病変部腸間膜脂肪組織(Fat Wrapping)におけるアディポネクチン遺伝子発現と蛋白発現をそれぞれ定量的PCRおよびELISA法にて定量したところ、正常コントロール群や他の腸炎群に比べて高値を示した。腸管に瘻孔を形成した症例ではアディポネクチン発現量が低かった。 2)マクロファージのサイトカイン産生に及ぼすアディポネクチンの影響(清原) IL-10のホモ欠損マウスから得たマクロファージのLPS刺激によるTNFαおよびIL-12産生に及ぼすアディポネクチンの影響を調べたところ、アディポネクチンはマクロファージのサイトカイン産生を著明に抑制した。 3)アディポネクチン欠損マウスにおける腸炎の検討(船橋、筒井) アディポネクチン欠損マウスにDSS腸炎を発症させ、その腸炎の程度を正常マウスの腸炎と比較検討した。まだ個体数が少なく十分なデータ解析はできていないが、現時点ではアディポネクチン欠損マウスと正常マウスの間で腸炎の程度に大きな差は認められていない。 4)アディポネクチン/IL-10二重欠損マウスの作成(筒井) アディポネクチンの腸炎抑制効果を検証するために、アディポネクチン/IL-10二重欠損マウスを作成した。IL-10のホモ欠損マウスは早期に腸炎を発症して弱るため、先ずアディポネクチンのホモ欠損マウスとIL-10のヘテロ欠損マウスを交配してアディポネクチン/IL-10の二重ヘテロ欠損マウスを得た。戻し交配を繰り返すことにより、アディポネクチンがホモ欠損、IL-10がヘテロ欠損のマウスを得た。現在このマウスを交配させてアディポネクチン/IL-10二重欠損マウス個体を作成中である。
