重症複合免疫不全症に対する安全性を高めた新しい遺伝子治療法の開発

研究概要	1.日本全国から依頼された重症免疫不全症患者の確定診断のために遺伝子診断を実施しており、これまでにX連鎖重症複合免疫不全症(X-SCID)をはじめ、Jak3欠損症(本邦第1例め)、IL-7受容体欠損症(アジア第1例め)、RAG1欠損症、Artemis欠損症を同定している。 2.変異蛋白が発現している重症免疫不全症に対する遺伝子治療を安全に施行するための簡易アッセイ系をX-SCID患者をモデル系として確立した。変異γc鎖が細胞表面に発現しているX-SCID患者に遺伝子治療を施行した場合、新たに遺伝子導入された正常なγc鎖が同じ細胞上でリガンドを競合しあう可能性が考えられる。そこで、細胞表面にγc鎖が発現していないT細胞株を用い、c-fosを遺伝子ターゲットとするトランジエントのアッセイ系を確立した。ただし感度はまだ十分とはいえず、感度を高めるのは今後の研究課題である。 3.自殺遺伝子を組み込んだγc鎖遺伝子発現ベクターを作成した。このベクターのバックボーンは既に遺伝子治療臨床研究で用いられているSFCMM3を使用した。自殺遺伝子は単純ヘルペス1型ウイルスのチミジンキナーゼである。今後、この発現ベクターをX-SCID患者から樹立したB細胞株、造血幹細胞や様々な血球系細胞に遺伝子導入する。この細胞を用いガンシクロビルによる細胞死の誘導を確認する。 4.倫理面での配慮 SCID患者の遺伝子診断については、倫理委員会にて承認されている説明文と同意書を用いインフォームドコンセントを得ている。
