研究概要	1.ラット肝細胞を用いた毛細胆管収縮運動観察の研究 (1)ラット肝細胞のcoupletを用いた研究 Coupletにより形成される毛細胆管収縮運動におけるサイトカイン(TNFα、IL-1)の影響について再現性を確認した。その上で胆汁酸およびCa濃度の影響を実施することができた。後者においては、Ca濃度については一定程度以上の濃度にて収縮運動抑制作用がみられたが、胆汁酸については実験設定範囲濃度内では明らかな傾向を確認できなかった。 (2)小型肝細胞を用いたin vitroで形成された毛細胆管運動上記タイトル内容については充分な確認をしえた。規則的運動ではないが、輸送路として伝搬に関わる運動として矛盾しないものと思われた。 2.肝細胞膜transporter分子発現の分子生物学的検討 Transporterとして複数個の因子を研究対象としたが、予備研究の結果最終的に2種にしぼった。そのうち研究開始計画でも焦点をあてていたMRP2の分析に関する安定性を確認しえたとともに治療を目的とした高圧酸素条件下の分析も成功した。 3.ヒト肝組織からの肝細胞分離平成16年度の研究としていた本研究を同時に実施し、ラットの小型肝細胞に類似した細胞の分離と同定、培養に成功した。しかし、明年よりその再現性を確認し、また短期間内の観察に終わっていることから今後の検討課題は多く残っている。
