研究概要

骨、軟骨での時計遺伝子群の発現レベルの日内変動を明らかにするために、48時間まで、夜昼の明暗条件および恒暗条件下で4時間ごとにラットを屠殺して、ラットの肋軟骨の成長板、静止軟骨および大腿骨からRNAを分離した。これらの組織においてPer, DEC1,DEC2の日内リズムが観察された。またこれらのRNAを用いて網羅的に日内リズム遺伝子を同定する実験を開始した。さらにDEC1発現アデノウイルスベクターを用いて、DEC1が軟骨分化を促進するのみならず、少し弱いものの骨芽細胞への分化も促進することを見いだして学会で発表した(分子生物学会シンポジウムなど)。さらにclock遺伝子の変異マウスの視交差上核では、DEC1,DEC2の日内リズムがほとんどなくなることを見いだした(Honma-S et al.2004 in press)。
一方、肝臓、筋肉では、DEC1とDEC2のどちらかの日内リズムがなくなったが、片方のリズムあるいは発現は維持された(能城ら、投稿準備中)。DEC1ノックアウトマウスの作成は順調である。またDEC1とDEC2の組織特異的トランスジェニックマウスのライン化も順調に進行しているので、来年度には解析が開始できる。
