電位依存性Na+チャネルβ-サブユニットの細胞接着分子としての新たな役割

研究概要	Na^+チャネルは、イオンチャネルを形成するα-サブユニット単独で、またはβ-サブユニットと会合しヘテロ2量体もしくは3量体として発現している。本研究ではβ-サブユニットの新たな生理機能を明らかにすることを目的とし、その発現調節機構おける蛋白リン酸化酵素の役割と、細胞分化の影響を解析した。ウシ副腎髄質細胞を無血清培地で培養すると、^3H-サキトキシンの結合を指標として測定した細胞表面のNa^+チャネル数が、濃度依存的かつ時間依存的に増加した。無血清培養により、MAPキナーゼファミリーのうち、ERKのリン酸化レベルが低下したが、p38とJNKのリン酸化レベルは変化しなかった。さらに、ERKの阻害薬であるPD98059とU0126を血清存在下の培養細胞に添加する、と、Na^+チャネル数が増加した。無血清培養またはERKの阻害薬によりα-サブユニットのmRNAレベルは増加したが、β1-サブユニットのmRNAレベルは変化しなかった。よって、α-サブユニットとβ1-サブユニットの発現調節機序は異なっており、ERKの抑制はα-サブユニットの発現のみを上昇させることが明らかとなった。一方、ラット褐色細胞腫PC12細胞はβ-サブユニットファミリーのうちβ1-とβ3-サブユニットを発現していた。神経成長因子(NGF)による神経分化に伴い、β1-サブユニットmRNAレベルは増加したがβ3-サブユニットmRNAは変化しなかった。以上の結果から、β-サブユニットの発現は神経分化によって調節されており、その細胞内機序はERKの活性化に非依存的であることが示唆された。
