東南アジアの生物から単離されたペプチド神経毒の合成研究

研究概要	1.新規イモ貝毒コノトキシンの合成とS-S架橋様式解明イモ貝(Conus marmoreus)から単離された新規ペプチド毒コノトキシンCMrII、CMrIII、CMrVの合成を行った。14残基のアミノ酸からなりC末端がアミド化されたCMrIIとCMrIIIの合成ではS-S架橋形成反応に高度希釈条件が必要であり、多大な労力を必要とした。一方、16残基のアミノ酸からなるCMrVの合成はスムーズに進行し、高収率で目的物を得た。得られた合成物はHPLCのco-injectionにより天然物と同一であることが確認された。CMrVを用いて部分還元とアルキル化、およびアミノ酸配列分析の組み合わせによるS-S架橋様式解明を試みた。その結果、3本の分子内S-S架橋の中でC3とC6の間に架橋があることが示された。続いてCysの保護基にAcm基とTrt基を使い分ける選択的2段階架橋法による合成物と天然物との比較を行い、C1-C5およびC2-C4の間にS-S架橋があることが示唆された。 2.サソリ毒のS-S架橋様式と立体構造および活性との相関アジア産黒サソリ(Heterometrus fulvipes)から単離されたHefutoxin 1の活性増強を目指して活性部位Lys19の近傍にある酸性のGlu16とGlu20を中性のAlaに置換したアナローグを合成した。合成はスムーズに進行し、置換部位はS-S架橋形成に重要ではないことが明らかとなった。得られたアナローグはいずれもKv1.2チャネルに対しては天然物より高い阻害活性を示したが、Kv1.1チャネルに対しては活性が低下した。このことから、16位と20位のアミノ酸はチャネルの選択性に寄与していると考えられた。その他の6カ所の塩基性あるいは酸性アミノ酸残基をAlaに置換したところ、S-S架橋形成はいずれもスムーズに進行した。
