魚類の即時的体液浸透圧調節機構に関する研究

研究概要	本研究では、体液よりも高張な海水から低張な淡水まで様々な浸透圧環境に生息している魚類の体液調節機構、とくにこれまで殆ど研究例の無い、環境浸透圧変化に迅速に対応する即時的適応機構、およびそれらの機構と従来から研究されてきた長期的適応機構との関連の解明を試みている。即時的な体液調節に関与する可能性がある分子としては、低分子ペプチドホルモンであるナトリウム利尿ペプチド(NP)ファミリー、グアニリン(GN)ファミリー、アドレノメデュリン(AM)ファミリーなどのホルモン群がある。平成15年度はこれらのホルモン群をコードするcDNAのクローニングから研究を開始した。NPファミリーについては、ウナギ、メダカ、ニジマス、トラフグ等からcDNAの探索を行った。その結果、魚類にはANP、BNP、VNP、CNP-1、CNP-2、CNP-3、CNP-4の7つの分子種が存在し(哺乳類には3種類しかない)、これらが単一の祖先分子から染色体の重複と遺伝子の縦列重複により生じたことを比較ゲノム解析により明らかにした。また、これらの分子をペプチド合成し、培養細胞で発現させた3種類の組換え受容体に対する活性を調べることにより、各分子が各々特異的に受容体を使い分けていることが明らかになった。GNファミリーについてはウナギからグアニリン、ウログアニリン、レノグアニリンの3種類の分子をクローニングし(哺乳類は2種類)、これらの分子の発現が海水中で増加していることを明らかにした。現在さらに2種類の受容体(哺乳類は1種類)がクローニングできつつある。AMファミリーについては、トラフグからAM1、AM2、AM3、AM4、AM5の5種類の分子をクローニングした。哺乳類ではこれまで1種類のアドレノメデュリン(AM1に相当)しか知られていなかったが、フグの情報をもとにして、最近哺乳類のAM2を発見することができた。
