汎用性医薬分子設計用ユニットに基づくヒスタミンH_3受容体特異的機能分子の創製

研究概要	従来より神経伝達物質ヒスタミンを内在性リガンドとする受容体としてH_1及びH_2受容体が知られており、そのアンタゴニストが抗アレルギー薬及び抗潰瘍薬として臨床利用されている。さらに、様々な薬理学的実験結果から第三のヒスタミン受容体としてH_3受容体の存在が示され、そのアゴニストあるいはアンタゴニストの医薬としての利用が期待されている。本研究は、シクロプロパン環の構造特性に着目して開発した医薬分子設計用ユニットを用いて、今までに報告例のないH_3受容体特異的機能分子、即ちH_1及びH_2受容体のみならずH_4受容体にも作用しないH_3アゴニスト及びアンタゴニストを創製するとともに、この医薬分子設計用シクロプロパンユニットの有用性を明らかにすることを目的として進めている。上記ユニットをもとに、種々のシクロプロパン型ヒスタミンの配座制限体を設計・合成し、4種のヒスタミン受容体サブタイプに対する親和性及び機能を調べた。その結果、folded型誘導体である(1S,2S)-2-(2-aminoethyl)-1-(1H-imidazol-4-yl)cyclopropane (AEIC)が期待したようなH_3特異的アゴニスト(K_i=4.3nM)であることが判った。この結果からヒスタミンがfolded型に類似した三次元構造でH_3受容体に結合することが示唆された。しかし、イミダゾール環とアミノ基の最適な空間的距離は現在のところ明らかでない。より親和性の高い化合物の創製を目的に、高活性を示したアゴニストAEICの構造をもとにして、ヒスタミンの薬理活性に必須と考えれるイミダゾールとアミノ基間の距離を種々変化させたfolded型誘導体3種を合成した。さらに、アゴニストからアンタゴニストへの機能変換を目的に、AEICのアミノ基に芳香環を導入した誘導体を数種合成した。これらは、H_3受容体結合部位周辺の脂溶性残基と導入した芳香環とが相互作用することで受容体の誘導適合が阻害されアンタゴニスト後にストとして機能することを期待して設計した。現在、これら新規に合成した化合物の薬理活性を検討している。
