腎特異的有機カチオントランスポータOCT2のプロモータ解析による発現制御機構解明

研究概要	本研究では、rOCT2の発現調節機構解明を目的として、rOCT2のプロモーター領域に注目し、テストステロンによるrOCT2発現調節機構の解析を実施した。 rOCT2転写開始部位上流の部分塩基配列をプローブとし、ラットゲノムライブラリーをスクリーニングしたところ、rOCT2のプロモーター領域に相当する約3kbのcDNA断片を得ることができた。この領域をpGL3 vectorに組み込み、アンドロゲンレセプターベクターとともにLLC-PK_1細胞に導入した後、テストステロンの影響を測定したところ、アンドロゲンレセプターが存在する場合にのみ、テストステロンによる転写活性の有意な上昇が確認できた。また、テストステロンによる転写活性の上昇には濃度依存性が認められ、雄性ラットの血中テストステロン濃度に対応する10nM以上の濃度では、誘導が飽和することが明らかになった。さらに、転写誘導に関わる塩基配列を特定することを目的として、rOCT2プロモーター領域の切断産物を作成し、プロモーター活性を測定したところ、-2250〜-1140位、並びに-3343〜-2250位の塩基配列を有する切断産物についてのみ、テストステロンによる転写活性の上昇が認められたことから、それらの領域に含まれる塩基配列に対してアンドロゲンレセプターが作用する可能性が示された。これらの結果から、rOCT2の腎発現性差並びに慢性腎不全モデルラットにおけるrOCT2発現低下には、アンドロゲンレセプターを介したテストステロンによるrOCT2の転写上昇が関与することが示唆された。
