結晶構造に基づくヘムオキシゲナーゼによるヘム分解機構の検討

研究概要	ヘムオキシグナーゼ(HO)は、基質であるヘムが自触的にO_2を活性化し、3段階の酸素添加反応(ヘム→α-ヒドロキシヘム→ベルドヘム→ビリベルジン)によってヘムを開環し、ビリベルジン・Fe・COを生成する。HOによるヘム分解の研究は過去数年間で著しく進展した。とりわけ第1の水酸化反応における活性酸素種がferric peroxideであることが明らかにされ、他のヘム酵素の触媒機構とは大きく異なることが知られた。我々は、HOの種々の結晶構造解析から、以下のことを明らかにした。第2反応では、電子要求性について内外の研究者間で議論されていたが、α-ヒドロキシヘムからベルドヘムへの転換がO_2のみで起こり、生成されたベルドヘムは2価であること、電子要求性についての研究者間の不一致は、結合体の調製方法と用いる還元系の違いによることを明らかにした。一方我々は、HO・ヘム結合型の結晶構造から、もっともヘム鉄に接近しているGly139、Gly143の主鎖が関与する第1反応のメカニズムを提唱し、アポ型の構造からヘムの結合が「誘導適合」されることを明らかにした。ヘム-N_3結合型の結晶構造から、第1反応においては、酸素原子間の結合はinversed heterolysisによって解裂されることを提唱した。CO結合型酵素の構造から、HOが目ら生成するCOによって反応阻害を受けない構造的基盤を明らかにし、HO-ビリベルジン鉄錯体の構造から、ビリベルジンがHOから遊離するメカニズムを推定した。
