mi突然変異マウスを用いたマスト細胞数をコントロールする因子の解析

研究概要	マウスmi遺伝子座は転写因子MITFをコードするが、このMITFに異常をきたしたmi/miマウスでは皮膚マスト細胞の数が3分の1に減少する。この原因としてマスト細胞の増殖因子stem cell factor(SCF)のレセプターであるKITの発現がmi/miマウスの皮膚マスト細胞で低下していることがあげられる。これまで用いたmi/miマウスは、C57BL/6系統であった(B6-mi/mi)が、戻し交配を行い、WBを遺伝学的背景とするmi/miマウスを作成した(WB-mi/mi)ところ、このWB-mi/miマウスではB6-mi/miマウスと異なり、皮膚マスト細胞数の減少がみられなかった。しかし、B6-mi/miマウス由来マスト細胞と同様、WB-mi/miマウス由来マスト細胞でもKITの発現が低下していた。このことは、KITの発現低下にも関わらず、皮膚マスト細胞の数を維持する何らかの因子がWB-mi/miマウスでは存在することを示す。N2世代のmi/miマウスでマッピングを行ったところ、ゲノム上のSCFをコードする領域近傍にこの因子が存在することがわかった。SCF蛋白の配列をB6系統とWB系統のマウスで比較したが有意な差はなかった。SCFには膜結合型と可溶型の2種類の蛋白があるが、後者の方が皮膚マスト細胞数に重要であることが報告されている。SCF mRNAにはalternative spliceによりlong form(SCF-L)とshort form(SCF-S)の2種類がある。可溶型SCFはSCF-L mRNAからの方が産生されやすい。そこで、B6系統とWB系統のマウスの皮膚におけるSCF-LとSCF-Sの量比を検討したところ、WB系統でSCF-L/SCF-S比が有意に高かった。以上のことより、WB系統のマウスではマスト細胞の数を増加させるSCFの量がB6系統のマウスよりも多く、そのためにKITの発現量の低下にもかかわらずマスト細胞の減少が認められなかったことが示唆された。
