肝再生医療に向けたオーバル細胞の発生・分化の解析

研究概要	ラットに2-アセチルアミノフルオレンを投与して部分肝切除を行なうと(AAF/PH)、門脈域に卵円形の核をもつオーバル細胞が出現する。オーバル細胞は肝細胞と胆管上皮細胞に分化できることから肝幹細胞由来と推定され、肝細胞の分裂・増殖が抑制された状況下で肝細胞の代わりに肝再生を担うと考えられている。一方、オンコスタチンM(OSM)はIL-6ファミリーのサイトカインであり、胎生期の肝細胞を機能的に成熟させる因子として知られている。近年、いくつかの因子が肝臓の発生と再生の両者に共通して働くことが示されているので、ラットのOSMとOSM受容体(OSMR)のcDNAを単離して、オーバル細胞におけるOSM受容体(OSMR)の発現とオーバル細胞株に対するOSMの影響を調べた。F344ラットにAAF/PHを施行してオーバル細胞の発生を促した肝臓では、OSMRの発現は実質細胞ではなくオーバル細胞を含む非実質細胞に認められ、OSMR mRNAのシグナルはオーバル細胞に一致した。更に、二重免疫組織化学染色法を用いて、サイトケラチン19の発現を指標にしてオーバル細胞と同定された細胞にOSMRが発現していることを確認した。また、オーバル細胞株をOSMで処理すると、増殖が抑制され、形態的に肝細胞様細胞へ変化し、肝細胞に特異的な酵素であるチロシンアミノトランスフェラーゼやトリプトファンオキシゲナーゼの発現が誘導されることが明らかになった。OSMは胎生期の肝細胞の成熟ばかりでなく、オーバル細胞による肝再生においても重要な役割を果たしている可能性がある。
