ストレス応答プロテアーゼ欠損変異株を用いたサルモネラワクチンの開発

研究概要

本年度の研究実施計画と成果を列挙する。
1.計画 ClpXP或いはLon欠損S.typhimuriumマウスへ感染させた各組織での感染菌の変化と組織から回収された菌の性状及び病原性を調べる(松井担当)。成果 in vitroではClpXP欠損S.typhimuriumは多鞭毛化し、Lon欠損S.typhimuriumはフィラメント化する。5x10^8CFUを6週齢の雌BALB/cマウスへ経口感染後、感染組織(パイエル坂、腸間膜リンパ節、脾臓)のホモジネートを一度平板に塗布し、生じたコロニーの菌の形態は感染前と同じく多鞭毛化とフィラメント化を呈していた。また、回収されたワクチン株はいずれも感染前と同じく弱毒株であった。以上の結果よりワクチン株は宿主内で強毒化する可能性は無いと考えられた。
2.計画 ClpXPと鞭毛形成遺伝子或いはLonとSulAの組み合わせで二重欠損変異株を作製し、菌の形態変化と病原性の関係を明らかにする(松井、関矢担当)。成果 いずれの二重欠損変異株も菌の形態変化(多鞭毛化とフィラメント化)は消失したが、マウスに対する病原性は弱毒化したままであった。従って、ClpXP欠或いはLon欠損S.typhimuriumとも菌体の形態変化と病原性は無関係であると言える。
3.計画 ClpXP或いはLon欠損S.typhimuriumは1回のみの経口投与でBAL/cマウスに強力な体液性免疫と細胞性免疫を誘導でき、強毒株からの攻撃を完全に防御できる。そこでこれらワクチン株により誘導される免疫応答を詳細に解析する(松井、江口担当)。成果 ワクチン株をBALB/cマウスに経口投与後の血清中の抗Salmonella LPS IgGのサブクラスと脾臓における各種サイトカインmRNAの発現量の推移から、ワクチン投与後、12週間はTh1タイプの細胞性免疫が感染防御に働いていると考えられた。一方体液性免疫はワクチン投与4週間後から誘導され、1年後でも感染防御に十分なレベルを維持していた。興味深いことにワクチン投与12週間はリステリアを皮下投与で攻撃しても感染を防御できた。また、粘膜でのIgAの産生が低下しているIL-5 KOマウスを免疫した場合も、通常マウスの免疫の場合と全く同じように強毒株の攻撃を防御できたことから、本ワクチンにより誘導される細胞性免疫が感染防御に重要な役割を担っていると考えられる。
