ラットモデルを用いたHTLV-I TAXの生体内での機能解析

研究概要	HTLV-Iによる腫瘍化にはウイルス蛋白Taxが深く関与していると考えられている。一方、ATL細胞の多くはTaxの発現か欠損していることも知られており、ATL発症過程におけるTaxの役割については不明な点も多い。本研究では、ラットHTLV-I感染T細胞にTaxを標的にしたsiRNAを導入し、TAXの発現低下が感染細胞に及ぼす影響をラットモデルで解析した。EGFP、およびTaxに対するヘアピンsiRNA発現ベクター(それぞれ、pEGFP-1/siEGFP、およびpEGFP-1/siTax)は、相補的合成オリゴヌクレオチトをアニールし、マウスU6 RNAプロモーターを組み込んだpEGFP-1に挿入することで作製した。はじめにpEGFP-1/siEGFP、およびpEGFP-1/siTaxの効果をマウスリンパ腫細胞株EL4より作製したEGFP発現細胞EL4/EGFP、およびEGFP-Tax融合蛋白発現細胞EL4/Gax細胞において解析し、本ベクターがEGFP、およびTaxの発現を抑制することを確認した。次にpEGFP-1/siTaxをラットHTLV-I感染T細胞株FPM1.BPに導入し、Taxの発現が低下しているサブクローン(FPMIB-siTax27)を得た。この細胞はコントロール細胞と比較してヌードラットでの腫瘍形成能、および転移能が著しく低下している一方で、Tax特異的CTLに対する感受性も低下していることが確認された。この結果は、TAXの発現抑制により、HTLV-I感染T細胞が生体内での増殖転移能を失うという感染細胞の生存に不利な変化を起こす一方で、宿主免疫系からのエスケープという有利な形質を獲得することを示すものと考えられる。本モデル系はATL発症過程での感染細胞の動態の一端を示している可能性が示唆され、ATL発症機序の解明、予防法の開発に有用であると考えられた。
