研究概要

Toll-like receptor 3 (TLR3)は線維芽細胞の細胞表面に存在し、ウイルスdsRNAの合成アナログであるpoly(I:C)を認識して、MyD88非依存的経路でタイプIインターフェロン(IFN-α/β)産生を誘導する。本研究では、TLR3に会合する新規アダプター分子、TIR-containing adaptor molecule-1 (TICAM-1)を同定し、TLRを介した新しいシグナル伝達経路を明らかにしたので報告する。TICAM-1は、712個のアミノ酸からなり、分子中央部にTIRドメインを、N末端側及びC末端側にそれぞれプロリンに富んだ領域を有する。TICAM-1をヒト培養細胞に過剰発現させると強力にIFN-βプロモーターを活性化し、IFN-β産生を誘導する。IFN-β遺伝子の転写調節領域には、NF-κB,AP-1 (ATF2/c-jun),IRF-3の結合サイトがあるが、TICAM-1の下流でNF-κBとAP-1が活性化されることをレポータージーンアッセイで確認した。また、TICAM-1の下流にIRF-3活性化経路が存在することを、HeLa細胞へのTICAM-1の過剰発現によるIRF-3のdimer形成の有無により明らかにした。最終的に、RNAi法により線維芽細胞やHeLa細胞のTICAM-1をノックダウンすることで、TICAM-1がTLR3を介したIFN-β産生に必須のアダプター分子であることを明らかにした(Nature Immunol.4:161,2003)。更に報告者らは、MyD88非依存的経路でおこる、LPS刺激によるTLR4を介したIFN-β産生に、TICAM-1が関与することを見出した。TICAM-1はTLR4とは直接結合しないことから、新規のアダプター分子が存在することが予測され、データベースサーチより第4番目のアダプター分子、TICAM-2を同定した(J.Biol.Chem.278:49751,2004)。Yeast two-hybrid、免疫沈降、レポータージーンアッセイ、RNAi法により、TLR4の下流では、TICAM-2とTICAM-1が協調して働き、IFN-β産生を誘導することが判明した。以上の結果より、TLRを介したシグナル伝達には、MyD88を介するNF-κB活性化経路とTICAM-1を介するIRF-3活性化経路が存在することが明らかとなった。
