消化管粘膜再生におけるTrefoil Factors(TFF)の分子生物学的解析

研究概要

胃粘膜,小腸粘膜及び結腸の単層培養細胞およびその欠損治癒過程追跡モデルを用いて、TFFの治癒過程への効果を検討し、EGF,TGFα,β,HGF,COX-2などの増殖因子またPPARなどのTFF発現に対する影響を検討した。さらに、細胞の悪性化の課程にTFFがどのように関与しているかを検討した。TFFのmRNA発現に関しては、pS2 geneのcDNAをRT-PCRを用いて作成した。それをプローブとしてNorthern blottingにより検討し、TFFのmRNA発現をPRlSM7700を用いて定量し、各種サイトカインのTFF産生に対する効果を検討した。胃粘膜の分化機能を有した胃癌細胞株MKN28、45,JR-Stなどを用いて、NSAIDのCOX-2発現、PG産生、PPARrの動態を検討し、それに与えるTFFの効果、さらにTFF産生との関連をmRNAレベルで解析した。その結果、NSAIDの胃粘膜に及ぼす効果に関して、COX-2を経由する以外に、TFFを経由するルートを見出した。これによって、NSAIDの胃粘膜細胞に対するいわゆる適応細胞保護作用のメカニズムが明らかになった。
TFFのTFF1,TFF2,TFF3などのsubtypeの存在に関して、各消化管には未知のTFFが存在している可能性が高く、これらをhomology検索によって同定、DNA配列を決定し、各疾患との関連性を検討した。消化管各種疾患のTFF発現をDNAアレイにより検討し、その臨床的意義を明確にした。NFkBにGFP蛋白遺伝子を導入しTFFの転写過程を解析している。さらに、免疫染色、ISHなどの方法を用いて、形態的遺伝子発現の研究を遂行した。
