ニコチンが肝機能(生理および病態)に及ぼす作用に関する研究

研究概要	喫煙が健康に及ぼす影響に関する研究は呼吸器疾患を中心として種々行われているが、肝臓に関しては殆ど行われていない。喫煙が肝臓に及ぼす影響を検討する目的でニコチン投与が肝血流に及ぼす効果をラットを用いて観察した。ウレタン麻酔下でウイスター系雄性ラットを用いて実験を行った。肝血流はレーザードップラーを用いて測定し、ニコチンを経静脈的および直接肝門脈に投与して肝血流の変化を観察した。その結果、ニコチンを全身性に静脈内投与すると血圧の上昇を伴う肝血流の増加が観察された。一方、ニコチンを肝門脈に直接投与すると血圧は変化を受けないが肝血流が減少した。ニコチン少量を前投与しておいて30分後に肝血流を減少させるニコチン量を門脈内に投与すると、反対に肝血流が増加した。以上の結果より、ニコチンは全身性に血圧を上昇させて肝血流を増加させるが、肝に対しては直接肝血流を減少させる。しかしながらニコチンの肝血流減少作用のメカニズムは不明である。また少量のニコチンの前投与によって2度目のニコチン投与による肝血流減少が逆に肝血流増加作用となることより、ニコチンによる肝のアダプティブ・サイトプロテクションの可能性が示唆された。
