研究概要

本研究の目的は、ファブリー病に対する遺伝子治療を確立するための基礎的検討を行うことである。ファブリー病モデルマウスにcDNA発現plasmidベクターにヒトα-galactosidase A_(Gal A)cDNAを接続した遺伝子を導入し、遺伝子治療を行う。導入遺伝子は、ヒトα-Gal A cDNA遺伝子を強力な発現力を示すCAGプロモーターを持つcDNA発現プラスミドpKSCX(ヒトCMV-IEエンハンサー、ニワトリβアクチンプロモーター、ウサギβ-グロビンポリ-A、カナマイシン耐性遺伝子)に接続して構築したpKSCX-α-Gal Aである。これを研究代表者が開発した方法(腎静脈へ注射する方法)(Hum. Gene Ther. 2002)を使って腎に導入し、治療効果について検討する。
1.遺伝子の導入方法の確立:研究代表者が開発したhydrodynamics-based transfectionによるラットの腎へのnaked plasmid DNAの導入方法を応用して、正常マウスの腎への遺伝子導入を検討した。正常マウスは、血中α-Gal A値が高いので、pKSCX-α-GaL Aを導入・発現させても、その効果を血中α Gal A値に反映させることは難しい。そこで、α-Gal A(101kDa)と同程度の分子量の蛋白を発現させられるか検討した。マウスの腎静脈は、ラットに比べて、細く短いが、全身麻酔下で、マウスを開腹し、左腎静脈と左腎動脈をクランプし、下大静脈と腹部大動脈との交通を遮断後、29Gのインスリン注射用シリンジを用いて、mouse IL-10/Fc(96kDa)発現plasmidベクターを導入、発現させることに成功した(Biochem. Biophy. Res. Commun. 2004)。
2.マウスの血液中、主要臓器中のα-galactosidase A濃度を分光蛍光光度計を用いて測定する方法を確立した。
3.10週齢のファブリー病モデルマウスの腎を電子顕微鏡で解析し、尿細管細胞にファブリー病患者と同様なGb_3の蓄積によるZebra bodyを認めた。
4.11週齢のファブリー病モデルマウスの腎にpKSCX-α-Gal Aを導入し、遺伝子治療の経過を観察中である。
