研究概要

1.インスリン抵抗性をきたすアンギオテンシンIIを用いた実験では、脂肪細胞に分化した3T3L1細胞では、IRS-1のセリンリン酸化は見られなかった。
2.インスリン抵抗性をきたすIL1による、IRS-1のセリンリン酸化を検討すると、JNKを介してセリン307、セリン612,636のリン酸化を認め、この時インスリンによる細胞内への糖の取り込み増加を抑制した。
3.3T3L1細胞で,anisomycin刺激の際に、IRS-1のセリンリン酸化が生じるがLDMからcytosolへの細胞内以降は生じない。SB600125はanisomycin刺激の際のLDMでのIRS-1のセリン燐酸化を強く抑制する。
4.Anisomycin刺激のによるIRS-1Ser612,Ser307燐酸化は、rapamycinによって、抑制される。
5.インスリン刺激によるIRS-1Ser612のセリン燐酸化は、SB600125,rapamycinで部分的に抑制され、両方で完全に抑制される。P70S6Kthr389燐酸化は、インスリン刺激の際には、SP600125で全く抑制されないが、anisomycin刺激の際には、SP600125で部分的に抑制される。
