膵β細胞の機能再生を応用した糖尿病治療への新たなる戦略

研究概要	糖尿病の大部分を占める2型糖尿病では、グルコース刺激に対する膵β細胞からのインスリン分泌能の低下が認められる。その成因の一つに膵β細胞内での糖代謝機構の障害による代謝因子の生成の減少が考えられる。近年になり、ミトコンドリア由来のglutamateが、インスリン分泌顆粒放出系に対し促進的に作用する代謝因子である可能性が示唆されている。そこで作用機序を明らかにする目的で、グルコースによるインスリン分泌機構に重要な役割を果たしているATP感受性K^+チャネル(K_<ATP>チャネル)活性に対する影響について、パッチクランプ法を用いて直接的な検討を加えた。まずglutamateの膜透過性アナログであるdimethylglutamateを用いて、パッチクランプ法のcell-attached modeによる検討を進めたところ、K_<ATP>チャネルの有意な抑制が認められた。そこでinside-outmodeを用いてglutamate自体によるチャネル活性への効果について検討を加えたところ、cell-attached modeの際とは異なりチャネル活性に対し何らの影響も認めなかった。以上の結果より、glutamateは直接K_<ATP>チャネルに作用してチャネル活性を抑制するのではなく、むしろ膵β細胞内でさらに代謝され、その結果生じる代謝因子がK_<ATP>チャネルを閉鎖してインスリン分泌顆粒の開口放出を促進するものと考えられた。またこの因子の候補の一つとして、現在のところATPの関与が最も考えられるが、今後glutamate代謝による細胞内でのATP分布の変化について詳細な検討を加える必要がある。
