不安障害の発症に関与する脳内機構についての分子生物学的研究

研究概要	FG7142を投与した雄性マウスの大脳皮質からtotal RNAを抽出し、その遺伝子発現量の変化について、cDNA microarray法でスクリーニングし、RT-PCR法を用いて定量的に調べた。成熟期では、c-fosを含めて14種類の遺伝子発現量が有意に増加していた。幼若期では、11種類の遺伝子は、FG7142投与による影響を受けなかったが、3種類の遺伝子は、変化の程度は小さい(+ 20%)が、有意に増加していた。また、成熟雄性マウスに拘束ストレス負荷と他の不安惹起物質(yohimbine)投与を行い、大脳皮質において、その発現量の変化をRT-PCR法で調べた。14遺伝子のうち、4種類の遺伝子が、FG7142の効果とほぼ同様に、その遺伝子発現量が増加することがわかった。このうち3種類は、前述した幼若期においてもFG7142投与によって発現量が増加する遺伝子であった。以上の結果から、2種類の不安惹起物質と拘束ストレスに共通して、その遺伝子発現量が増加する遺伝子が、4種類ほど明らかになった。これらは、いずれも、FG7142による変化が成熟期で顕著になることから、生後発達過程で形成される不安の神経回路に関与している可能性があり、こういった神経回路の脆弱性が不安障害の発症に関与していると考えられた。一方、FG 7142を投与した雄性マウスの大脳皮質から抽出したtotal RNAを用いて、RAP-PCR法と電気泳動法によって、未知の遺伝子を検索したところ、その遺伝子発現量がFG7142投与によって変化する3種類の遺伝子が見つかり、現在クローニングを進めている。
