加齢黄斑変性症に対する新しい治療法の開発

研究概要

zinc finger型転写因子であるIntestinal Kruppel-like Factor(IKLF)は下肢虚血モデルの血管新生、心臓血管リモデリングに関与する事が分かっているが、眼球における作用は不明である。今回、まず8から12週齢のIKLFヘテロ欠損マウスの眼球切片を作成し、網膜血管構造の異常の有無を観察した。また、IKLFヘテロ欠損型および野生型マウスを生後7日目(P7)からP12まで75%酸素下で飼育後、通常酸素下に戻す事でROPモデルを作成し、眼内病的血管新生へのIKLFの関与を検討した。その結果IKLFヘテロ欠損型マウスでは網膜および血管構造に異常をみとめず、ROPモデルでは網膜の無血管領域の面積には有意な差を認めなかったが、新生血管領域面積は74%(P=0.0317)減少していた。以上より、KLFヘテロ欠損マウスでは網膜血管に異常は観察されないが、IKLFはROP発生において抑制的に作用することが示唆された。
