P2X受容体発現細胞を用いた脳組織細胞外ATP放出イメージング法の開発

研究概要

脳組織において本当にATPは細胞内から細胞外へ放出されているのか?この仮説をイメージングによって検証することが本研究の目的である。まずP2X2受容体を発現させ準HEK293T細胞が広領域のATP変化を検出し得るか検討した。高い空間分解能を得るために、HEK293Tをconfluentに蒔きP2X2受容体を発現させ細胞内Ca^<2+>濃度もしくは膜電流を計測したところ、ATPに対する応答は長く持続的で、時間的な分解能が著しく低いことがわかった。また、発現効率も細胞間でかなり差があり十分な空間分解能は得られなかった。これらの問題の克服は来年度の課題である。一方、この研究期間中に、細胞外ATPの検出において2つの大きな進歩があった。ひとつは、初代脳由来培養細胞で超高感度光子検出カメラとluciferin-luciferase反応を用いて細胞外ATPが検出されたとする報告、もうひとつは、バイオセンサーを用いた脳組織内局所的ATP濃度のリアルタイム計測の報告である。後者の開発者Nicolas Dale博士との共同研究を申し入れたが、2004年度まではイギリス国内でしか使用できないという特許の規定である、との返事であり、それ以降に共同研究を進める計画を立てている。超高感度光子検出カメラ(貸与)を用いて、海馬、大脳皮質、孤束核スライスにおけるATP放出のイメージングを試みたところ、ATP放出を画像化し得た。電気刺激による細胞外ATP濃度上昇は、(1)刺激電極の近傍に限局し、軸索終末近傍では観察されない、(2)テトロドトキシンによって消失しない、そして、(3)高K+濃度刺激によっては生じないことが明らかになった。これらは、脳スライスで電気刺激によってATPが放出されるとする現在までの報告の多くに重大な疑義を投げかける重要な結果である。
