細菌性サイトカインの精製と応用に関する研究

研究概要

細菌性食中毒が発生すると、原因菌を患者から分離するとともに、原因食品を特定するために一般的な細菌学的検査が行われるが、2割程度しか原因食品は特定されてこなかった。この原因の一端は、細菌が食品中でVNC(Viable but Nonculturable)状態、即ち生きてはいるが培養不能な状態に陥っているからで、そのため検出率が極端に低下していると我々は考えている。近年海水や河川中の環境下や食品中における種々の細菌のVNC状態への変化が報告され、我々は腸管出血性大腸菌やサルモネラ、リステリアなどの病原細菌が容易にVNC状態に変化する現象を観察してきた。すなわちVNC状態への移行は、環境のストレスに応答して、自己を防衛するために細菌が自ら保有する生命維持機構であるといえる。しかし、VNC状態の病原細菌は現在の細菌検査手法では分離培養不可能であるにも関わらず、動物に病原性を保持していることが知られている。VNC状態の細菌は栄養豊富な場で必ずしも蘇生するわけではなく、逆に栄養不足状態の方が蘇生しやすいことが判っている。このことからVNC状態の細菌は生体内で栄養が豊富になり蘇生するというよりも、何らかの蘇生誘導物質により培養可能な状態に戻り(蘇生し)、病原性を発現すると我々は仮説をたてている。実際我々は細菌の培養濾液中にVNC細菌の蘇生に関与する物質(細菌性サイトカイン)が存在することを現象として確認している。このことから細菌間で共通に産生される細菌性サイトカイン類似物質がVNC細菌の蘇生を誘導したり、感染後宿主側が類似物質を産生しているのかもしれない。そこで、このサイトカインの同定と応用を本研究で実施する。本年度は、この遺伝子を同定し、蘇生に関与することを見出した。
