薬物依存形成に関与する分子群の簡便な検索法の確立および応用

研究概要	我が国において、最も乱用されている依存性薬物はメタンフェタミンであるが、薬物依存を引き起こすメカニズムについては、ほとんど分かっていない。小生は、平成12年度から5年間の予定で文科省科学技術振興調整費による研究助成を受け『依存性薬物により誘発される精神障害の機構の解明の研究』をテーマに研究班を組織し、10名の薬物依存研究者を統括している。今までに本研究班からいくつかの薬物依存関連遺伝子を見出してきた。それらの遺伝子を見つけるためには、依存モデル動物を用いて脳内での発現変化や行動薬理学的な検討を1つ1つ行っていかなければならない。多くの依存関連遺伝子候補タンパクをスクリーニングするためには、時間、費用およびマンパワーにおいて不可能に近いとされている。当萌芽研究では、薬物依存に関連する蛋白のスクリーニングを可能にするを目的として、培養細胞を用いた薬物依存モデルの確立を目指している。メタンフェタミンの作用の1つに、細胞間隙でのドパミン遊離量の増大がある。ドパミン遊離量が増大することによって、さまざまな精神毒性が引き起こされていると考えられている。メタンフェタミン添加によってドパミン遊離の増大が観察される細胞系を確立することができれば、薬物依存関連遺伝子のスクリーニングに活用することが出来ると考えられる。そこで、本年度は、メタンフェタミン刺激でドパミン遊離が増大する細胞の確立を目指した。モデル細胞として、SH-SY5Y細胞を用いた。この細胞は、ドパミン産生能を有し、ドパミン受容体も発現している。この細胞にメタンフェタミンを作用させ、培養上清中のドパミン遊離量を測定したが、検出限界以下であった。培養上清の濃縮や測定系の改善を試みたが、ドパミンを検出することが出来なかった。そこで、この細胞と同じ特性を有する上、ドパミントランスポーターも発現していることも報告されているPC12細胞を用いて、検討を行ったところ、メタンフェタミン添加時のみ、培養上清中のドパミンの測定が可能であった。次年度は、メタンフェタミン添加量とドパミン遊離量の用量依存性を確認すると共に、このモデル細胞に、我々がすでに見出している薬物依存関連タンパクの添加や遺伝子を導入し、動物実験で得ているデーターと一致する結果が得られるかどうかを確かめる予定である。
