がん細胞パネルと計算機による抗がん剤の3次元ファーマコフォア抽出

研究概要	1.化合物の作用機作別分類がん細胞パネル評価で得たフィンガープリント情報(作用機作を反映)により化合物をクラスタリングし作用機作別に分類した。その中でTopoisomerase I (Topo I)を阻害する複数化合物(リガンド)のファーマコフォア解析を試みた。 2.自動配座解析プログラムCAMDAS(分担者広野ら開発)による配座集団生成 CAMDASプログラムは、高温分子動力学計算に基づきサンプリングした配座についてエネルギー極小化後、2面角の値によるクラスタリングでエネルギー極小配座の代表集団を自動生成する。Topo Iを阻害する5種のリガンド(PDC,FM-02,acetomycin,NF-1173,LGZK-69)の配座集団生成を行った。得られた配座数はPDC:10950:、FM-02:1、acetomycin:41、NF-1173:1568、LGZK-69:5665であった。 3.重ね合わせプログラムSUPERPOSE(分担者広野ら開発)による結合配座決定 SUPERPOSEでは、リガンド内の疎水性、水素結合供与/受容性の原子団ごとに特性球を置き、分子を特性球で表現する。三次元空間で異なる化合物の配座同士を重ね合わせ、同じ性質の特性球が重なったら加点、異なる性質の特性球が重なったら減点することで、高得点で重なる配座を抽出し、同一特性の原子団が最も良く重なる配座の組み合わせを求める。Topo I阻害剤であるtopotecanが結合したTopo IのX線解析による座標(PDB:1K4T)からtopotecanを抜き出し、これに対し上記2で得られた配座集団をSUPERPOSEで重ね合わせ、各リガンドの結合配座を推定した。各リガンドから最良の重ね合わせとなる待合配座をTopo Iのtopotecan結合位置に置くことで、各リガンドの結合様式が得られることになる。現在進行中である。
