SAGE同定新規胃癌関連遺伝子に対するaptamerによる組織局在と増殖制御

研究概要

SAGE(serial analysis of gene expression)法により、胃癌について網羅的遺伝子発現を行い、胃癌と非癌部胃粘膜とのSAGEライブラリーの比較から、胃癌で特異的に高発現する様々な遺伝子を同定した。その内、カルシウム依存性レクチンスーパーファミリーに属するregenerating gene type IV(REGIV)は、遺伝子導入系を用いた機能解析から、分泌蛋白をコードすること、アポトーシスを抑制すること等を見い出した。そこで、SELEX(systematic evolution of ligands by exponential enrichment)法により、RNA aptamerの精製を試みた。まず、RT反応およびPCRにより全長cDNAを調整し、これを6xHisタグした後に、発現ベクター(pCE-30UA)に組み込み、大腸菌で大量培養した。得られた6xHisタグ標識蛋白は、金属キレート・アフィニティークロマトグラフィーを用いて精製した。次に、両端にfixed sequenceを持ち中央部に40塩基のランダムsequenceを含むオリゴヌクレオチドライブラリー(1micro-moleスケール当り10^<14>種類程度)をsolid-phase synthesizerにて10^<14>種類程度で作成した。これをstarting poolとし、合成したREGIV蛋白と反応させた後、ニトロセルロースフィルターで結合したオリゴヌクレオチドをfixed sequenceをプライマーとしてPCRで増幅、この結合、分別、増幅のサイクルを10回以上繰り返した。特異的結合は、ゲルシフトアッセイで確認中である。特異的かつ親和性の高いオリゴヌクレオチドが得られ次第、PCRで増幅しaptamerとする予定である。
