自己寛容をコントロールする新しい転写因子FOXP3の胸腺における機能の解明

研究概要

scurfy(sf)の形質は、sf胸腺により教育された自己反応性T細胞によって生じていると考えられる結果(Godfrey, V. et al. Proc.Natl.Acad.Sci.USA 88:5528-32,1991)が発表されていた。本研究の1つの目的はsf胸腺によるT細胞分化異常を明らかにすることであった。sf胸腺上皮細胞に異常があるとすれば、Foxp3の発現が無いために生じる遺伝子発現の異常に起因するものである。そこで野生型(C57BL/6;B6)胸腺上皮細胞株(国立感染症研究所笠井博士より供与)数種を用いてFoxp3の発現について検索した。その紳果、調べたいずれのクローンにおいてもFoxp3の増幅バンドは検出されなかった。すなわち、Foxp3が胸腺上皮に発現しないことが、sf胸腺による寛容誘導異常に直接関連するか否かは現在のところ不明である。このことを証明するためには、sf胸腺移植実験による自己免疫誘導実験が必要であるが、マウスの供給不良と動物施設のマウス肝炎ウイルスによる長期間の閉鎖によって未了である。
マクロファージ遊走阻止因子(MIF)は、最初に生物活性が明らかとなったサイトカインである。MIFはグルココルチコイドにより発現増強するなど、免疫寛容・抑制などとの関連が推測された。MIFと調節性T細胞機能の関連を解析する目的で、マクロファージ遊走阻止因子(MIF)トランスジェニックマウス(Tgm)由来のCD4^+25^+T細胞の抑制活性を調べた。野生型(B6)の(CD25T細胞を反応細胞として抗CD3抗体とX線照射B6脾細胞を培養する系に一定の比で、野生型・TgmのCD4^+25^+T細胞を添加して増殖抑制能を比較したところ、Tgm由来CD4^+25^+T細胞の抑制活性がやや低下していた。GITRに関しては発現が増加しており、抑制活性低下と相関しているものと考えられた。
