薬毒物の作用機序の解析に有用な新しい質量分析法

研究概要

覚醒剤をはじめとする乱用薬物は、神経系のドーパミンレセプター等の受容体に作用し、下流の情報伝達系に作用すると考えられており、種々のタンパクがリン酸化等の修飾を受けると考えられている。また、リンやヒ素のような無機薬毒物は、タンパクのSH基に結合して毒性を発揮すると考えられている。これら修飾タンパクの解析に関し、多くの方法が応用されている。しかし、近年は超微量の目的分子を検出することが求められて、従来の方法では対応しきれなくなっているのが現状である。しかし、薬毒物の作用機序をプロテオミクスから明らかにする場合、従来法よりイオン化効率が高く、よりソフトなイオン化法を用いた新しい質量分析法の開発が望まれるが、現在、ESI及びMALDIを越えるイオン化法はまだなく、発想の転換が求められている。今回我々は修飾ペプチド解析のための新しい方法論の開発を目指したい。
まず、我々は、レーザースプレー-飛行時間型(LS-TOF)質量分析計(MS)を用いて、リン酸化ペプチドの高感度検出及び定量法の確立を試みる。その際、モデルとして、チロシン水酸化酵素(TH)のN末端部分のペプチドを用いる予定である。その理由としては、当酵素がカテコールアミン系の律速段階の酵素であり、覚醒剤やコカイン等の乱用薬物の影響を受けやすいこと、かつN末端にリン酸化部位が多数あり、複数個のリン酸化修飾の解析モデルとして有効であると考えられることが挙げられる。THのリン酸化部位は、主に19,31,40番目のセリン残基であると考えられている。まず、19番目のセリン残基を含む20mer程度のペプチドを合成し、リン酸化ペプチドと非リン酸化ペプチドについてLC上での分離条件の検討を行い、次いでLC-TOF-MSによるイオン化効率を検討する。このとき、レーザーの出力、移動相の検討と共に、正・負イオンの両モードで検討し、最適なイオン化効率を検討する。
