造血幹細胞の可視化とその応用

研究概要

造血幹細胞と血管内皮細胞に特異的に発現するマウスEndomucin遺伝子とTMTSP遺伝子の開始コドンにin-frameにVenus遺伝子を挿入したノックインマウスの作成を進行中である。すでに、TMTSPの開始コドンにin-frameにVenus遺伝子を挿入したノックインマウスが得られた。Endomucin遺伝子に関してはまだ進行中であるが、今後、ノックインマウスの骨髄の切片を詳細に解析し、骨髄における造血幹細胞ニッシェの解析を行う。また、5-FU投与等による骨髄抑制を行った後の骨髄回復期にニッシェがどのように変化するのか、造血幹細胞の自己複製がどのように起こるのかを観察する。また、TMTSPノックインES細胞、および抗Endomucin抗体を用いて、ES細胞からの血液細胞の発生につき検討を行い、TMTSPとEndomucinがともに血液細胞発生のマーカー分子となることを確認した。今後、これら2つの分子を用いて、さらに血液細胞の発生の機序につき明らかにしていきたい。
