神経幹細胞への染色体部位特異的遺伝子組込み法の開発

研究概要	(1)Cre-loxP法によりRepの発現を制御したAAVS1特異的遺伝子組込みベクターを構築した。まず、HSV-tkプロモーターとRepとの間に、loxP配列で挟まれた転写終結シグナルを含むstuffer配列を挿入した。このstuffer配列にITR配列を連結したマーカー発現ユニットを逆向きに配置した。このRep発現誘導カセットを導入した細胞にCre発現ベクターを感染させ、Repが一過性に発現することを確認した。さらに、この切り出されたstuffer配列中のITRにRepが結合することによって、第19番染色体AAVS1領域に遺伝子組込みが生ずることが期待される。HeLa細胞を用いて、Souther blot解析によるAAVS1領域の遺伝子再構成を検討し、マーカー遺伝子のAAVS1領域特異的遺伝子組込みを確認中である。さらに、ITR配列を連結したマーカー発現ユニットとRep発現誘導カセットを有するアデノウイルスベクターを構築中である。 (2)アデノウイルスベクターの感染には細胞表面上の受容体であるCoxsackie adenovirus receptor (CAR)に結合することが必要であるが、細胞の種類によってはCARの発現が低いために遺伝子導入効率が不十分である。この対策として、アデノウイルスと未分化な細胞との接着機構に着目し、これを応用した幹細胞でのアデノウイルスベクター発現システムを開発した。未分化細胞に効率良く感染させるため、アデノウイルス受容体とc-KitのリガンドであるSCFをHisタグとリンカーで連結した融合アダプター分子を作成した。これを用いて、アデノウイルスベクターの造血系細胞における感染増強効果をFACSにて検討中である。
