DNAチップを用いた口腔微生物叢の網羅的定量的プロファイリング

研究概要

本年度は、1.DNAチップの基本となる口腔細菌特異的プローブの設計および作成、2.口腔試料を用いて、DNAチップ用プローブと既存のPCRプライマーの比較検討、従来法による口腔細菌叢の解析を行い、試料の一部は将来のDNAチップ解析用試料として保存した。
1.歯垢細菌特異的プローブの設計および作成
(1)プローブの設計
口腔細菌叢構成細菌の内、約30菌種の16S ribosomal RNA塩基配列(GenBankより)をもとに、DNAチップ用の細菌特異的プローブ(オリゴDNA)を設計した。
(2)プローブの作成
(1)の設計をもとに、DNAチップ作成共同研究企業へプローブの作成の検討を依頼した。DNAチップへの搭載の可否を現在検討中である。
2.従来法による歯垢細菌叢の解析
(1)口腔試料の採取
う蝕病巣、歯周病巣からのプラーク試料を口腔試料として採取し、一部は、将来のDNAチップ解析用試料として凍結保存した。試料の採取は、学内倫理委員会規程に則って行った。
(2)菌種特異的PCR法による菌種の検索
(1)の試料を用いて、ミュータンスレンサ球菌などの病原微生物を、既存のプライマーおよびDNAチップ用プローブの塩基配列を持つプライマーを用いて検出し、その感度や特性を比較した。両者には大きな違いはなかった。
(3)培養法による菌種の検索
試料を通法にて血液寒天平板で培養し、コロニー数から細菌叢全数を求め、各コロニーの微生物種をPCR-RFLP解析から同定した。この結果は、将来得られるDNAチップ解析結果と比較検討を行う予定である。
