研究概要

気管支喘息およびアトピー性疾患は20世紀後半、先進諸国において約10倍増加し、現在、対数的に増えつつあると報告されている(BMJ 316:607-16,1998)。それに伴う医療費の増加や患者本人および家族への負担は深刻であり、その原因の究明と予防法の確立は急務の課題である。気管支喘息は気道の炎症がその本態と考えられているが、気道炎症のバイオマーカーとなるものはいまだ存在しない。本研究は喘息のバイオマーカーとなりうる遺伝子およびたんぱく質をプロテオミクスとGeneChipの技術により同定する。
喘息患者発作時および同じ患者の回復期の血漿を用いてプロテインチップおよび2次元電気泳動による解析をおこなった。発作時と回復期のサンプルの収集は困難であったが現時点で12検体を収集した。プロテインチィップおよび2次元電気泳動で数百の蛋白が分離できるが、現在までに得られたデータでは発作時と回復期で差の認められるたんぱく質は同定されていない。平成16年度はさらに感度の高い方法を用い、サンプル数を増やして解析をおこなう予定である。
喘息およびアトピー性疾患の遺伝子解析としては血清ECP値を調節する多型(Am J Respir Crit Care Med, 2003)、Toll-like receptorの機能的変異(Clin Exp Allergy, 2004)、アトピー性皮膚炎関連遺伝子(Gene and Immunity, 2003)等の研究発表を行った。
