軟体動物の嗅覚学習に伴う脳内情報表現の可塑性と行動変化の解析

研究概要	はじめにナメクジ前脳ノンバースティングニューロン選択的にカルシウム感受性色素を負荷する方法を確立した.さまざまな条件で染色を行い比較検討した結果,触角-脳標本を作成し,前脳の内層のみにおいてシースを除去した後,カルシウム感受性色素(rhod-2 AM)の溶液中でインキュベートすることにより,ノンバースティングニューロンの選択的染色を行うことができることが明らかになった.このようにして染色した標本を冷却CCDカメラを用いて光学測定することにより,数十個のノンバースティングニューロンの活動を同時に,単一ニューロンの解像度で測定することが可能になった.測定の結果,無刺激時にはノンバースティングニューロンはほとんど活動していないこと,触角神経束の単発電気刺激により,一過的にカルシウム濃度の上昇を生じることが明らかになった.これはバースティングニューロンが無刺激時にも常に周期的に興奮を繰り返していることと対照的である.そこで次に触角に匂い刺激を与えたところ,一部のノンバースティングニューロンでカルシウム濃度の上昇が見られた.匂い刺激に対する応答は,ほとんどの細胞で一過的なものであったが,一部の細胞では約1Hzのカルシウム振動が数周期にわたって生じた.また一部のニューロンでは,ある匂いには反応し別の匂いには反応しないという,匂い刺激の種類による特異性が見られ,前脳ノンバースティングニューロンが匂いの種類をコードしていることが示唆された.
