小胞体の機能と制御のダイナミクス

研究概要	小胞体の機能と制御に重要な役割を果たす転写誘導機構としてATF6経路とIRE1-XBP1経路をこれまでに同定している。ATF6はプロテオリシスを受けて活性化される小胞体膜結合性転写因子である。ATF6切断酵素がゴルジ装置に存在するため、ATF6は小胞体ストレスに応答して小胞体からゴルジ装置へ移行した後切断され、その結果膜から遊離した細胞質側断片が核へ移行して転写を活性化する。まず、セリンプロアテアーゼ阻害剤AEBSFが小胞体ストレスに応答したATF6の切断を阻害するが、ATF6の小胞体からゴルジ装置への移行には影響を及ぼさないことを明らかにし、AEBSFがATF6活性化機構解明の上で重要なツールとなることを示した。また、小胞体ストレス下で高次構造が異常になった分泌タンパク質や膜タンパク質は小胞体に留められる一方で、ATF6と正しい高次構造を形成しているタンパク質は選別されてゴルジ装置へと運ばれることを明らかにし、小胞体の高い選別能を証明した。さらに、ATF6がSREBPによって媒介される脂質新生を阻害することを見いだした。IRE1-XBP1経路においては、転写因子XBP1をコードするmRNAが小胞体ストレス下IRE1依存的にフレームスイッチ型スプライシングを受けて活性型のpXBP1(S)が産生される。酵母のXBP1カウンターパートであるHAC1に自律的発現量調節機構が備えられていることを見いだし、この機構が酵母からヒトまで保存されていることを明らかにした。C型肝炎ウィルスがIRE1-XBP1経路を阻害することによって、自身の分解を妨げていることを見いだした。糖尿病を発症するアキタマウス由来の膵臓β細胞では、ATF6経路とIRE1-XBP1経路がともに活性化されており、小胞体ストレスが糖尿病の発症に関与する可能性が示唆された。このように小胞体の機能と制御に関する多くの知見を得た。
