Whether the Tom40 channel, which comprise of β-barrel, can be opened laterally has been a hot topic for the past decades. We provided proofs for this possibility. In vitro, we observed fusion proteins with cytochrome b2 holding Om45 pathway targeting signal from the IMS, followed by DHFR for preventing the fusion proteins to go through the Om45 pathway, were inserted into the OM and left the Tom40 channel gradually. In vivo, we observed growth recover from yeast expressing fusion proteins with matrix targeted non-stop protein holding the targeting signal of Om45 pathway, which suggests that non-stop fusion protein does not occupy the Tom40 channel. Proving the possibility of the lateral gating of the Tom40 channel has great impact on understanding the import of mitochondrial proteins.
