The original intent of the proposal was to develop an imaging platform that can correlate proteome information with metabolome information during a 2-year research term. In the first year of the study, a hybrid fluorescence-Raman imaging system that is capable of correlating protein expression patterns with metabolome fluctuation information was successfully developed and applied to biological studies involving stem cell differentiation.
In this second year, I have published the study on Scientific Reports, and further improved the photobleaching problem that has prevented time-lapse imaging using the hybrid imaging method by carefully choosing the laser excitation condition. Although the original plan was to use bioluminescent probes to reduce photobleaching problem, it was found out that the substrate for bioluminescent probes also photobleach during laser irradiation, thus I was forced to change the research direction. Fortunately, by carefully tuning the incident laser intensity and accumulation time to obtain Raman spectral images, I was able to perform time-lapse hybrid fluorescence-Raman measurements over the necrotic process of cells. I am now collaborating with a medical lab to monitor the cytochrome c redox state using the hybrid imaging method and we are producing interesting results. The work will be continue on even though the term of this research fund is due.
Together with the journal publication of the hybrid flourescence-Raman imaging work, I have also presented the study in 4 domestic conferences this year, thanks to the support of this research fund.
