研究実績の概要 |
This project is an extension of the JSPS Kaken WakateB, of which we aimed to isolate novel cellulolytic microbes from wood-devastating insect. Thus, achievements can be divided into two; 1) Isolation and characterization of wood-devastating insect symbionts, 2) Combinatorial enzyme assays to optimize polysaccharide degrading enzymes. In the former, we finished isolations of several bacteria and fungal species from Xiphydria wood wasp from Hokkaido forest. We are in the process of submitting this study to Scientific paper. Combinatorial enzyme assay project has been ongoing. I spent a month at the U.S. department of energy - Joint Genome Institute, to communicate with a host scientist, Dr. Yoshikuni, and his colleagues. The enzyme assay plat form is ready to be operated this year.

理由
We have finished isolations of several bacteria and fungal species from Xiphydria wood wasp, and we have completed biochemical and proteomic analyses of the culture supernatant from the fungus, Daldinia recipients sp. soita. I will soon submit the manuscript to the scientific journal to complete this project. For the combinatorial enzyme study, I have been focusing on testing a kelp deconstruction by using a glycoside hydrolases (GHs) and pectate lyases (PLs). The first screening of enzymes by a combinatorial assay was completed, and good candidates from each enzyme families, GHs and PLs, were selected. Between two enzymes, end products from kelp hydrolysis were determined by a solid state NMR. In this year, we will determine kinetics of kelp hydrolysis by combination of two enzymes.

今後の研究の推進方策 |
We might proceed the insect-symbiont microbe study depending on the capacity of my laboratory personnel who might be interested in to carry out this project further. For example, we are interested in to search for insects that convert wood materials into nutrients. For the combinatorial enzyme study, we will push forward this project by completing all the assays for target enzymes. Then, I will start preparing the manuscript to wrap up this project.
