研究概要

ケモカインレセプターCCR4を標的としたT細胞系腫瘍の分子解明と分子標的治療の研究を進めてきた。結果、T細胞系腫瘍における両分子の発現解析として、成人T細胞性リンパ腫/白血病(ATLL)、peripheral T-cell lymphoma-unspecified、Anaplastic large cell lymphoma(ALK陰性)において高率にCCR4が高率に発現するとともに、予後不良因子であること、また、in vitro、in vivoにおいて抗CCR4抗体はADCCを介した抗腫瘍効果を発揮することを見出した。ATLL腫瘍細胞に発現する表面分子として、CD4,CD25陽性分画にはCCR4が発現すること、制御性T細胞のマーカーであるFOXP3が共発現することを見出しATLL細胞の細胞起源としての制御性T細胞の可能性を強く示した。ホジキンリンパ腫においてホジキン細胞にCCR4を発現する細胞株を検出し、in vitroでの抗CCR4抗体による抗腫瘍効果を確認した。皮膚原発T細胞性リンパ腫についても、in vitro及びin vivo (SCIDマウス)での抗CCR4抗体による抗腫瘍効果を確認した。ホジキンリンパ腫生検組織においてCCR4とFOXP3の二重染色によりホジキン細胞(Reed-Sternberg細胞)周囲の反応性リンパ球は制御性T細胞が集簇していることを示した。以上に結果より、CCR4分子はT細胞系腫瘍、さらにはホジキン病において重要な標的分子であるとともに、腫瘍免疫学的なその関与を示し、今後の研究領域をさらに拡大することができた。また、T細胞性腫瘍に対するヒト化抗CCR4抗体療法の第I相試験を実施し、13例に安全に投与できることが明らかになった。
