研究概要

遺伝子操作による人為的分化転換の可能性、特に、非造血系組織が造血系へ分化転換することが可能かどうかを検討した。具体的には、骨格筋細胞を脱分化させる作用のあるMsx1遺伝子を、アデノ随伴ウイルス(AAV)ベクターを用いてマウス骨格筋で一過性に発現させ、そこに出現する単核細胞が造血系への分化能を有するかどうかを調べた。AAVベクターで導入した遺伝子は、その殆どが染色体に組み込まれない形で存在するため、骨格筋細胞がMsx1の働きにより脱分化して増殖するようになると、Msx1遺伝子は次第に失われ、細胞は再び分化を始めるという想定に基づくものである。脱分化誘導用としてマウスMsx1遺伝子、コントロールベクター用としてマウスEpo遺伝子をAAVベクターに搭載し、また非発現AAVベクターも用いた。マウス前脛骨筋にAAV-Msx1あるいはコントロールベクターを注射し、同部位の骨格筋より分離した単核細胞を用いてin vitroコロニー形成実験を行ったところ、Msx1ベクターを筋注した部位で造血前駆細胞の増加を認めた。さらに、同部位の単核細胞は、in vivo造血系再構築能を有する未分化造血幹細胞を多く含むことを見出した。この骨格筋の中に出現する造血幹細胞が骨格筋の脱分化・再分化という現象に基づくものか、あるいは組織障害を修復するために移動してきた骨髄由来のものであるかを調べるため、造血系と骨格筋を区別できるようにしたキメラマウスを作製し、研究をさらに進めている。その他、AAVベクターで骨格筋に導入した遺伝子の動態解析をin vivoイメージング装置を用いて行った。Msx1遺伝子を導入した場合は、コントロール遺伝子と異なり、数週後には失われていく傾向を認め、予想した結果が得られた。
骨格筋から脱分化段階を経て造血幹細胞を生み出すことが可能となれば、新たな幹細胞ソースになるものと期待される。
