研究概要

【目的】Wntシグナルの伝達因子であるβ-カテニンは細胞接着や転写調節に関与し、発がんとの関連が注目されている。一方、成人T細胞白血病(ATL)の原因レトロウイルスであるヒトT細胞白血病ウイルスI型(HTLV-I)のトランスフォーミング蛋白質Taxは種々の細胞生存シグナルを活性化することで細胞を不死化するが、Wntシグナルへの関与は不明である。本研究ではHTLV-Iの発がん機構におけるβ-カテニンの関与について検討した。
【結果】HTLV-I感染細胞株ではβ-カテニン蛋白質の発現はTaxの発現と相関しており、β-カテニン蛋白質は核に蓄積していた。また、β-カテニン依存性のTcf転写活性をレポーターアッセイで解析したところ、Tax発現感染細胞株で高い傾向にあった。Tax非発現感染細胞株では、プロテアソーム阻害剤処理にてβ-カテニン蛋白質の蓄積が観察されたが、Tax発現感染細胞株ではβ-カテニン蛋白質のさらなる蓄積はみられないことから、Tax発現感染細胞株ではβ-カテニン蛋白質が安定化していると考えられた。野生型TaxやNF-κB経路を活性化できないTax変異体M22の発現プラスミドをHeLa細胞に導入すると、β-カテニン蛋白質の発現、β-カテニン蛋白質の安定化およびβ-カテニン依存性のTcf転写活性の誘導が認められた。しかし、CREB経路を活性化できないTax変異体703およびK88Aでは上記の活性は誘導されなかった。また、Taxにより誘導されるβ-カテニン依存性のTcf転写活性はCREBの優性抑制変位体により阻害された。β-カテニン分解を促進するGSK-3βはPI3K-Akt経路により不活化されることが知られている。HTLV-I感染細胞株におけるAktの活性化はβ-カテニンやTaxの発現と相関しており、野生型TaxやM22をHeLa細胞に導入するとAktの活性化が認められ、GSK-3βが不活化されたが、703やK88AではAktを活性化できなかった。PI3K阻害剤でHTLV-I感染細胞株を処理すると、Akt活性が阻害され、β-カテニンの発現が減弱し、細胞生存率が低下した。また、Taxにより誘導されるβ-カテニン依存性のTcf転写活性もAktの優性抑制変異体の導入により阻害された。
【総括】TaxはCREB経路の活性化を介してAktを活性化し、GSK-3βを不活化する。その結果、β-カテニンの分解が抑制され、β-カテニン依存性のTcf転写活性を増強し、細胞を不死化すると考えられた。
