Mutational robustness in ribozyme quasispecies

Importance: Viral populations characteristically form ensembles of genotypes that quickly diversify from a founding genotype under the selection pressures. Mutant genotypes often have no deleterious effects on the fitness, conferring mutational robustness. The role of mutational robustness in the survival of viral population, which circumvent antiviral therapies has not been quantified. This research quantifies the role of mutational robustness on the survival of populations of ligase ribozyme evolving in the presence of manganese mutagenic agent.

Findings: Evolution without mutagen approached extinction in approximately 42 generations, whereas with manganese there was no extinction. Genotypic characterization shows more genotypes generated during the evolution of mutagenic treatments, but that have accumulate no more than 5-10 mutations each. Genotype networks show lower extinction rates under mutagenic treatment. Mutational robustness, estimated from the degree distribution, shows that the mutagenic treatment has four genotypes with more than 50 connections, and seven genotypes with more than 10 connections. Whereas, non-mutagenic population has one genotype with more than 50 connections, and three with more than 10 connections.

Fitness values measured for common genotypes in each enzymatic step were added to the networks to quantify the presence and extent of neutral networks. The manuscript resulting from this research is in preparation and submission process.

There has been few methodological challenges, but overall the results are ready for publication.

今後の研究の推進方策

I am in the process of adding the fitness values to the genotypic networks, as traits, to observe if clones with similar fitness values tend to cluster together forming neutral networks. In addition, I am making measurements of mutation rates and the effect for the mutagen on the population size. I am also preparing the manuscript for submission.

Populations under further mutation rates are being developed such that we can further quantify mutational robustness and characterize the trend effect of the mutational rate on the development of these populations. Once the populations are developed, genotypic characterization and genotypic networks will be done, using the experimental pipeline developed. The results from the research of the second year are expected to produced another publication.

I will need to buy some extra reagents this fiscal year, so I let some money to be carried over for the specific purchase (digital PCR reagents) that I had not considered in the beginning.
