109 neuropeptides and peptide hormone transcripts were recovered from the transcriptome of the acorn barnacle Megabalanus rosa. These include some known neuropeptides such as Allatostatins, Sulfakinin, FMRFamide family neuropeptides, pigment dispersal hormone and insulin like peptides. While the majority of these peptide hormones have been implicated in adult functions and behaviors, our result for the first time reveal their expression in barnacle cyprid larvae nervous system. By three color IHC experiments, the whole nervous system (anti-acetylated tubulin with red 2nd antibody), the location of SIFamide, Allatostatin and FMRFamide neuropeptides (anti-neuropeptide antibody with green 2nd antibodies) and larval anatomy (DAPI, blue) are simultaneously illustrated by three colors immunohistochemistry experiments. We further extend our sequence data to compare homologous neuropeptide sequences from other barnacle species and decapod crustaceans, through which we discover high degree of conservation. In particular, SIFamide exhibited similar spatial expression pattern in M.rosa and a distant clade the parasitic barnacle Sacculina yatsui. In M.rosa, SIFamide express not only in the major neural fiber, but also in distinct location of the larval body. Our results enable further study on the commonality of nervous system among different barnacle clades and therefore have major implication on the evolution of barnacle.
