研究概要

フィーダーフリーのTT2 ES細胞を樹立した。ICRのモルラ胚との凝集法でキメラ作製が可能である。また、C57BL/6由来のES細胞の樹立し、100%キメラが得られた。しかし、生殖系列への伝達率は、TT2に比較し悪い。トランスジェニック及びキメラマウス作製を、計244件実施した。ES細胞相同組換えに関する相談は、17件にのぼった。その内容は、ベクター構築に関してパーツとなるプラスミドや部位特異的組換えに関するプラズミドの分与、feeder free ES細胞の分与、ベクター構築(アームの長さや場所等)の相談、ES細胞培養技術指導、ES細胞へのベクター導入とクローン単離、キメラマウス樹立およびマウス交配指導、トラップES細胞を利用した遺伝子置換のベクター構築の指導等であった。37機関から361件の微生物学的モニタリングの依頼があり、4608検体の検査を行った。マウス肝炎ウイルスの感染はほとんど見られなくなった。海外へのマウスの送付が増加しているため、英語での検査成績依頼が増加している。リボゾーマルRNA遺伝子は13箇所に別れて遺伝子座が存在する。このような場合、トラップベクターが挿入されたリボゾーマル遺伝子の位置を容易に特定できない。そこで、交配による遺伝子マッピングを行い、18番または19番染色体上にあることを、明らかにした。遺伝子操作マウスの寄託凍結保存数は767件、供給系統数は950件であった。MHVに感染したマウス等500系統ののクリーニングを行った。生殖工学に関する技術指導を国内では4回20名、国外では中国北京大学で5名について行った。生殖工学の技術を網羅した英語のCD版を作製した。凍結精子の体外受精用の培地を開発した。CARD R-BASEから、EGTC、JMSR、FIMRE、IMSRへのリンクを張り、より利用しやすい環境を作った。
