研究概要

遺伝性ラット(Tsc2およびBhd変異体)腎発がんの研究から、その過程で発見されたERC遺伝子が、ヒト「アスベスト・中皮腫」の診断・腫瘍マーカーの開発に進み、抗体治療を含めた、新規治療法の開発へと展開した。さらに、ヒト結節性硬化症の原因遺伝子産物であるtuberin(Tsc2産物)は、ラパマイシン感受性mTOR経路を抑制する機能を持つ。この経路の遮断による結節性硬化症の臨床治験が試みられているが、必ずしも著効を示す結果は得られていない。我々はtuberinの機能の全体像を明らかにした。新たな治療・予防法の標的となる経路の解明に資する。
