研究概要

本研究計画では「生命システムの破綻」の表現型としての臨床情報を日常臨床の中から網羅的・体系的に収集することと、ゲノム研究、疾患メカニズム研究の知見を統合することによって、生命・疾患をシステム的に理解し、それに基づいて個々人への最適で、安全かつ効率的な医療を提供することを目指した。東京大学医学部附属病院において、ゲノム-臨床情報統合データベースシステムの構築と運用を実現した。このデータベースを駆使して生活習慣病発症進行と相関する遺伝子多型を同定した。これまでわれわれが行ってきた分子生物学的検討で心血管病に重大な影響をもたらすことが解明されてきた因子、例えば転写因子KLF5のプロモーター領域のSNPが心血管病発症と有意相関することを確認し、分子生物学的検討でそのSNPが有する生物学的意義を解明した。ゲノム情報とデータベースに時系列表示された臨床情報とを統合することにより疾患の遺伝的危険因子を同定し、さらに疾患メカニズム解析研究まで発展させることができた。
