研究概要

自己免疫性甲状腺疾患の感受性遺伝子を連鎖解析により探索し、Zn-fingerとAT-hookドメインを有する転写因子、ZFATを新たに同定した。さらに、ZFATがサイトカインや抗原受容体構成分子をはじめとする免疫応答に関連した一群の遺伝子発現を抑制していることを見出した。ZFATノックアウトマウスは胎生致死となり、初期発生に重要であることが示された。ヘテロ欠損マウスでは、二次リンパ組織に於ける細胞数増多、抗原受容体刺激によるB細胞の増殖亢進、移植片対宿主病応答の遅延がみられ、ZFATがB細胞の増殖ならびにTh1/Th2バランスを制御する可能性が示唆された。
