研究概要

1)特異的な神経伝達系の傷害に対しての神経移植療法によるシナプス機能再生過程を評価するためパーキンソン病モデル動物に対する胎児ドーパミンニューロン移植実験を継続して行なった。パーキンソンモデルラットのPET画像にて廃絶していたトランスポーターが細胞移植4週間後に増加していく様子を同一個体で追跡し、ドーパミントランスポーター機能によって画像化されるシナプス前機能と、D2受容体計測により明らかにされるシナプス後機能評価を平行して行った。同一個体で複数の計測を経時的にできるPETならではの実験法である。
中型動物用PET計測に加え研究期間内に新たに導入された小型動物用のmicro-PETの稼働を開始し、さらに解剖学的な解像度の増した実験系で研究を継続した。
2)同じ実験系を用い神経細胞移植による行動変化の定量的評価を加えることで神経移植の効果を明らかにし、一個体での経時的な動物PETによる記録と対比し、行動と受容体計測の定量的評価を対比した。複数の論文として発表した
3)全脳に分布する神経細胞の損傷時の機能評価を可能とするための受容体計測法の開発を行なった。パーカッションモデルラットによる神経外傷モデルを作成し、オートラジオグラフィー法を用いた神経受容体計測と高次脳機行動解析(open-field、T-maze test、water maze)による神経伝達異常に関する検討を開始した。受傷部位を中心に広汎に広がるグリア反応を描出できる末梢性ベンゾジアゼピン受容体計測をPETにて行った(論文準備中)
4)ラットや脳外科手術で得られたヒト脳のスライスを培養液内で神経活動を保った状態で神経刺激を加えその神経反応伝播を光学計測法とポジトロン標式リガンドによるブドウ糖代謝計測を組み合わせて解析する新しい手法を確立し後者に関して論文発表するとともにデーターを蓄積した。
